چکیده

Neuroprotective properties of ketosis may be related to the upregulation of hypoxia inducible factor (HIF)-1alpha, a primary constituent associated with hypoxic angiogenesis and a regulator of neuroprotective responses. The rationale that the utilization of ketones by the brain results in elevation of intracellular succinate, a known inhibitor of prolyl hydroxylase (the enzyme responsible for the degradation of HIF-1alpha) was deemed as a potential mechanism of ketosis on the upregulation of HIF-1alpha. The neuroprotective effect of diet-induced ketosis (3 weeks of feeding a ketogenic diet), as pretreatment, on infarct volume, after reversible middle cerebral artery occlusion (MCAO), and the upregulation of HIF-1alpha were investigated. The effect of beta-hydroxybutyrate (BHB), as a pretreatment, via intraventricular infusion (4 days of infusion before stroke) was also investigated following MCAO. Levels of HIF-1alpha and Bcl-2 (anti-apoptotic protein) proteins and succinate content were measured. A 55% or 70% reduction in infarct volume was observed with BHB infusion or diet-induced ketosis, respectively. The levels of HIF-1alpha and Bcl-2 proteins increased threefold with diet-induced ketosis; BHB infusions also resulted in increases in these proteins. As hypothesized, succinate content increased by 55% with diet-induced ketosis and fourfold with BHB infusion. In conclusion, the biochemical link between ketosis and the stabilization of HIF-1alpha is through the elevation of succinate, and both HIF-1alpha stabilization and Bcl-2 upregulation play a role in ketone-induced neuroprotection in the brain.

Neuroprotective effects of crocin on the histopathological alterations following brain ischemia-reperfusion injury in rat

Objective(s): Some histopathological alterations take place in the ischemic regions following brain ischemia. Recent studies have demonstrated some neuroprotective roles of crocin in different models of experimental cerebral ischemia. Here, we investigated the probable neuroprotective effects of crocin on the brain infarction and histopathological changes after transient model of focal cerebral...

Effect of pentoxifylline on brain edema in a rat model of transient focal cerebral ischemia

Pervious studies have shown that pentoxifylline (PTX) has beneficial effects in reduction of stroke and brain trauma injuries in experimental animals. However, there is very little and controversial information about the effect of PTX on brain edema in cerebral ischemia. Therefore, the aim of this study was to determine the effects of different doses of PTX on brain edema and neurological m...

L-NAME and 7-Nitroindazole Reduces Brain Injuries in Transient Focal Cerebral Ischemia in Rat

Background: The role of nitric oxide (NO) of endothelial or neuronal origins in cerebral ischemia and reperfusion injuries are far from being settled, extending from being important to not having any role at all.  Objective: To investigate the role of NO of endothelial and neuronal origins in ischemia/reperfusion injuries in focal cerebral ischemia, L-NAME, a non selective NO synthase inhibitor...

Inhibition of Angiotensin-Converting Enzyme Reduces Cerebral Infarction Size in Experimental-Induced Focal Cerebral Ischemia in the Rat

Background: The role of Renin Angiotensin System (RAS) in ischemic/reperfusion (I/R) injuries is not fully elucidated. Furthermore, it is not clear whether inhibition of RAS by Angiotensin-Converting Enzyme (ACE) inhibitors has beneficial effects in terms of protecting the brain from I/R injuries. In this study enalapril is used as an ACE inhibitor to evaluate the role of RAS in I/R injuries in...

Quantitative evaluation of Blood Brain Barrier permeability in transient focal cerebral ischemia in the rat

Introduction: Development of brain edema following focal cerebral ischemia exacerbates primary ischemic injury. Blood brain barrier (BBB) opening is an important part of edema named as vasogenic brain edema. In this study, quantitative alterations of BBB permeability is experimentally evaluated using transient focal cerebral ischemia in the rat. Methods: Two groups of male rats (ischemic and sh...
